ABSTRACT

Background & objectives: Timely intervention is needed to minimize the economic losses of vector-borne bovine anaplasmosis which can be possible by the isothermal amplification assay. Methods: Anaplasma marginale in the cattle of south Gujarat, India was detected in the PCR and LAMP by amplifying the fragment of msp5 gene. The PCR product was digested with EcoRI, and sequenced to confirm its pathogen specific detection. Results: Species specific PCR observed a band of 457 bp of msp5 DNA following 1% agarose gel electrophoresis. Positive LAMP reaction turned into yellow colour while negative sample depicted original pink colour. A detection limit of PCR and LAMP was up to 10-6 and 10-8 of the original genomic DNA of A. marginale, respectively. A single cut site of EcoRI was observed in the PCR product. Current msp5 DNA sequences of A. marginale (MW538962 and MW538961) showed 100% homology with the published sequences. Monophyletic lineage type relationship was observed with high bootstrap proportion among the msp5 DNA sequences of A. marginale in the phylogram. Prevalence rate of A. marginale was significantly higher (p<0.05) in the PCR [43/280 (15.36%)] and LAMP [62/280 (22.14%)] than the microscopic technique [17/280 (6.07%)]. Diagnostic sensitivity, specificity, positive and negative predictive values at 95% CI for LAMP assay with respect to PCR were 93.02%, 90.72%, 64.52% and 98.62%, respectively. Interpretation & conclusion: Thus LAMP can be a practical alternative to the PCR for the diagnosis of A. marginale infection in the cattle even in field condition

ABSTRACT

@#Anaplasma marginale is the most prevalent tick-borne haemoparasite of cattle and causes huge economic losses to the dairy industry worldwide. This study aimed to determine the occurrence of A. marginale infection in blood and tick samples collected from livestock animals in the districts located in Khyber Pakhtunkhwa (KPK), Pakistan. A total of 184 blood and 370 tick samples were included in this study. It has never been reported that sheep, goats, and cattle in Tank, Ghulam Khan, Birmil and Miran Shah areas were infected with A. marginale. All samples of blood and ticks were collected through random sampling from March 2021 to January 2022 from cattle, sheep and goats and screened through PCR for anaplasmosis by using primer pairs of Anaplasma spp. Three hundred and seventy ticks were collected from infested hosts (120/184, 64.21%). Among the four morphologically identified tick species, the highest occurrence was recorded for Rhipicephalus sanguineus (n=138, 37.29%), followed by Rhipicephalus microplus (n=131, 35.4%), Rhipicephalus annulatus (n=40, 10.81%), Hyalomma anatolicum (n=31, 8.37%), and Hyalomma marginatum (n=30, 8.1%). The occurrence of female tick was highest (n=160, 43.24%), followed by nymphs (n=140, 37.38%) and males ticks (n=70, 18.9%). Among these ticks, A. marginale was detected in female ticks of R. microplus, and R. sanguineus. Molecular identification of A. marginale was confirmed in 120 out of 184 blood samples and 6 out of 74 tick samples. Overall, occurrence of A. marginale in blood and tick samples was found to be 65.21% and 8.1% respectively. Species-wise occurrence in blood samples of goats were 71.11% followed by sheep 68.31% and cattle 50%. Specie-wise occurrence of A. marginale in tick samples of cattle were 12.5% followed by goats 6.89%. The obtained sequence showed similarity with A. marginale reported from Kenya and USA. We report the first PCR based detection of A. marginale infection in blood samples and in R. sanguineus ticks of goats simultaneously.

Resumen Históricamente, la especie bovina ha sido de vital importancia para el hombre como animal de abasto, por lo que el cuidado y protección a esta especie es muy importante. A nivel mundial existen diversas enfermedades que atacan el ganado bovino, entre las cuales se encuentra anaplasmosis bovina. La presente investigación no experimental de campo tiene como objetivo evaluar la dinámica de anticuerpos anti-Anaplasma y determinar en qué momento el animal presenta infección activa. Para ello, se escogió una población de 35 becerras ubicadas en la unidad de producción del municipio Crespo, en el estado de Lara, Venezuela. A las becerras, de 0 a 7 días de nacidas, se le extrajo la sangre en tubos con anticoagulante, para realizar frotis de capa blanca y determinación del hematocrito, y sin anticoagulante, para la inmunofluorescencia indirecta. Posteriormente, fueron muestreadas cada 15 días hasta obtener un total de 12 muestras por cada becerra. Para analizar los datos, fue utilizada la prueba estadística X2 del programa estadístico Epi Info versión 3.5.4. Se obtuvo un 79,05 % de positividad y 100 % de seropositividad para Anaplasma marginale. Se concluyó que los altos valores obtenidos con las pruebas serológicas son indicativos de animales con memoria inmunológica, lo que quiere decir que no necesariamente estarían infectados al momento del muestreo. Asimismo, se confirma la capacidad de la especie bovina de crear un estado de premunición ante la anaplasmosis a edades tempranas y mantenerla por un periodo prolongado, de modo que se encuentra una estabilidad enzoótica en la unidad de producción.


Abstract Bovine species has been historically of high importance to the human beings as a provision animal. Therefore, it is highly important to care and protect this species. There are different diseases attacking the bovines around the world, such as the bovine anaplasmosis. This non-experimental research in the field aims to evaluate the dynamics of anti-Anaplasma antibodies and to determine when the animal has an active infection. To do so, a population of 35 female calves was used from the production unit in the town Crespo, Lara State, Venezuela. Female calves, from 0 to 7 days born, were taken blood samples in test tubes with anticoagulant. Then a white smear test was carried out and the hematocrit was determined. Samples without anticoagulant were taken to do an indirect immunofluorescence test. The calves were taken blood samples every 15 days in order to complete 12 samples from each individual calf. For the data analysis, the statistical test X2 from the software Epi Info v.3.5.4 was used. Positive tests accounted for 79.05 % of the samples and seropositiveness for Anaplasma marginale reached 100%. It is concluded that the high values resulting from the serologic tests indicate that these animals have an immune memory, which means, in turn, that they were not necessarily infected at the time of being taken the samples. Likewise, this study confirms that this species is able to create a pre-immunity condition to the anaplasmosis at an early age and to keep it for a long term. This way, they reach an enzootic balance in the production unit.

Abstract Although anemia has been historically linked to Haemonchus contortus infection, other infectious agents, such as hemotropic mycoplasmas and tick-borne disease pathogens, may also lead to anemic crisis in sheep. This study has aimed to investigate infections related to anemia in a sheep herd from Bandeirantes City, Paraná State, southern Brazil. Seven out of forty-two (16.6%; 95% CI: 8.32-30.6%) sheep were positive for hemoplasmas by a PCR targeting the 16S rRNA gene and all tested negative for A. marginale/A. ovis and Babesia/Theileria spp. by PCR based on msp4 and 18S rRNA genes, respectively. Two (4.7%; 95% CI: 1.32-15.79%) animals were infested with Rhipicephalus microplus ticks. Fecal egg counting was performed in 38 sheep and 24 (63.15%; 95% CI: 47.2-76.6%) presented > 500 eggs per gram. Phylogenetic analysis of partial sequences of the detected hemotropic Mycoplasma sp. 16S and 23S rRNA genes confirmed that the animals were infected with Mycoplasma ovis. Polymorphism analysis of partial 16S rRNA sequences showed three different genotypes of M. ovis infecting sheep assessed in the present study. Mycoplasma ovis and gastrointestinal nematodes occurs in sheep from the northern region of Paraná State.


Resumo Embora a principal causa de anemia seja historicamente relacionada à infecção por Haemonchus contortus, outros agentes infecciosos, como micoplasmas hemotrópicos e patógenos transmitidos por carrapatos, também podem causar quadros anêmicos em ovinos. O presente estudo objetivou investigar infecções relacionadas à anemia em um rebanho de ovinos, na cidade de Bandeirantes, Estado do Paraná, sul do Brasil. Sete (16,6%; 95% CI: 8,32-30,6%) de 42 ovinos foram positivos para hemoplasmas pela PCR do gene 16S rRNA, enquanto todos foram negativos para A. marginale/A. ovis e Babesia/Theileria spp. por ensaios da PCR baseados nos genes msp4 e 18S rRNA, respectivamente. Dois (4,7%; 95% CI: 1,32-15,79%) animais estavam infestados por carrapatos Rhipicephalus microplus. Dos 38 animais nos quais foi realizada a contagem de ovos por grama de fezes (OPG), 24 (63,15%; 95% CI: 47,2-76,6%) apresentaram valores >500 para OPG. A análise filogenética das sequências parciais dos genes 16S rRNA e 23S rRNA de hemoplasmas confirmou a infecção por Mycoplasma ovis. A análise de polimorfismos de um fragmento do gene 16S rRNA mostrou a ocorrência de três genótipos diferentes de M. ovis nos animais. Mycoplasma ovis e nematódeos gastrointestinais ocorrem em ovinos da região nordeste do Estado do Paraná.

ABSTRACT

The study aimed to evaluate and compare the clinical, laboratory and pathological aspects of buffalo and bovine experimentally infected with AmRio 2 strain of Anaplasma marginale. Four Murrah buffaloes and four crossbred cattle were used in the experiment, which two animals of each species were splenectomized. Strain AmRio 2 of A. marginale was inoculated in all experimental animals. Clinical exams, Packed Cell Volume (PCV), blood counts, blood smears, rickettsemia, necropsy and histopathology were performed in all cases. Semi-Nested-PCR (snPCR) for the msp5 and snPCR for the msp1α target gene for identification of A. marginale in blood samples from animals was done. From positive samples for msp1α snPCR, samples were analyzed for the amino acid sequences of this gene. Two splenectomized cattle presented apathy, pale mucous membranes, jaundice, hyperthermia, and severe anemia. The remaining experimental animals did not show clinical signs. The rickettsemia in all animals was less than 1%. The mean PCV of the splenectomized cattle was below 20% at two-time points after infection. On the blood count, the main changes were observed in splenectomized calves and were characterized by a decrease in red blood cells, hemoglobin, PCV and platelets (p <0.05). All animals presented leukocyte elevation by increased lymphocytes, however, with no significant difference. The average prepatent period was two days in all the animals. The average incubation period in cattle that became ill was 25.5 days, and death occurred, on average, 63 days after inoculation of the strain. The necropsy findings were characterized by pale carcass, ascites, enlarged liver, distended gallbladder, and thick bile. Histopathological findings included infiltration of macrophages and lymphocytes in various organs, hepatic sinusoidal dilatation, and necrosis of the large intestine. In snPCR for the msp5 gene, 100% of the animals were positive in at least one evaluation. And in the snPCR for the infection of the msp1α target gene was also found in all animals in at least one sample evaluated. However, sequencing revealed only five animals, including the bovine which died, with a similarity of the amino acid sequences with AmRio 2 strain of A. marginale. It is concluded that the splenectomized cattle died due to anaplasmosis caused by the inoculated strain and the buffalo were more resistant compared to cattle. Buffaloes can be an alternative to cattle rearing in areas with a high occurrence of clinical cases of anaplasmosis.(AU)


O estudo teve como objetivo avaliar e comparar os aspectos clínicos, laboratoriais e patológicos de búfalos e bovinos infectados experimentalmente com estirpe AmRio 2 de Anaplasma marginale. Para isso, foram utilizados quatro bubalinos Murrah e quatro bovinos mestiços, sendo dois animais de cada espécie, esplenectomizados. Estirpe AmRio 2 de A. marginale foi inoculada em todos os animais. Foram realizados exames clínicos, hematócrito, hemograma, esfregaço sanguíneo com avaliação de riquetsemia, necropsia e histopatologia, além de, Semi-Nested-PCR (snPCR) para o gene alvo msp5 e snPCR para o gene alvo msp1α para identificação de A. marginale nas amostras de sangue dos ruminantes. A partir das amostras positivas na snPCR msp1α, foram selecionadas amostras para análise das sequências de aminoácidos deste gene. Dois bovinos esplenectomizados apresentaram apatia, mucosas pálidas, icterícia, hipertermia e anemia severa. O restante dos animais não apresentou sintomatologia clínica. A riquetsemia em todos os animais foi menor que 1%. A média do hematócrito dos bovinos esplenectomizados esteve abaixo de 20% em dois momentos após infecção. Ao hemograma, as principais alterações observadas foram nos bovinos esplenectomizados e caracterizaram-se por redução de hemácias, hemoglobina, hematócrito e plaquetas (p<0,05). Todos os animais apresentaram elevação de leucócitos por aumento de linfócitos, porém, sem diferença significativa. O período pré-patente médio foi de dois dias em todos os animais. O período de incubação médio nos bovinos que adoeceram foi de 25,5 dias e estes morreram em média 63 dias após inoculação da estirpe. Os achados de necropsia caracterizaram-se por carcaça pálida, ascite, aumento de volume do fígado, vesícula biliar distendida e bile espessa. À histopatologia, verificou-se infiltração de macrófagos e linfócitos em diversos órgãos, dilatação dos sinusoides hepáticos e necrose do intestino grosso. A snPCR para o gene msp5, revelou 100% dos animais positivos em pelo menos um momento de avaliação. E na snPCR para o gene alvo msp1α também verificou-se infecção em todos os animais em pelo menos uma amostra avaliada. Entretanto, o sequenciamento revelou apenas cinco animais, incluindo os bovinos que morreram, com similaridade das sequências de aminoácidos com estirpe AmRio 2 de A. marginale. Conclui-se que os bovinos esplenectomizados morreram em virtude de anaplasmose provocada pela estirpe inoculada e os bubalinos foram mais resistentes em comparação aos bovinos. Finalmente, os búfalos podem ser uma alternativa à criação de bovinos em áreas com alta ocorrência de casos clínicos de anaplasmose.(AU)

ABSTRACT

Anaplasma marginale and A. platys were detected and characterized (16S rDNA sequence analysis) from dairy and indigenous cattle, and the latter in domestic dogs in Vietnam. A phylogenetic tree was inferred from 26 representative strains/species of Anaplasma spp. including 10 new sequences from Vietnam. Seven of our Vietnamese sequences fell into the clade of A. marginale and 3 into A. platys, with strong nodal support of 99 and 90%, respectively. Low genetic distances (0.2–0.4%) within each species supported the identification. Anaplasma platys is able to infect humans. Our discovery of this species in cattle and domestic dogs raises considerable concern about zoonotic transmission in Vietnam. Further systematic investigations are needed to gain data for Anaplasma spp. and members of Anaplasmataceae in animal hosts, vectors and humans across Vietnam.

Abstract Anaplasma marginale and piroplasm species are widespread among Brazilian cattle herds. Both of these tick-borne pathogens hamper livestock production and cause a significant economic impact. Although buffaloes have demonstrated a high level of adaptability, data on tick-borne pathogens are scarcely reported in Brazil. Thus, the aim of this study was to screen water buffaloes from the state of Maranhão for piroplasm and A. marginale occurrence using PCR assays. All samples were negative for A. marginale. One of the 287 (0.35%) water buffaloes tested was positive for Theileria sp. Sequencing of the 18S rDNA fragment (356 bp) showed that the Theileria sp. identified was closely related to the T. buffeli /orientalis group. Future studies on the clinical signs of infection and the main vector in this country are needed.


Resumo Anaplasma marginale e espécies de piroplasma são amplamente distribuídas no rebanho bovino brasileiro. Ambos os patógenos transmitidos por carrapatos dificultam a produção pecuária e causam um impacto econômico significativo. Embora os búfalos tenham demonstrado um alto nível de adaptabilidade, dados sobre patógenos transmitidos por carrapatos são raramente relatados no Brasil. Assim, o objetivo deste estudo foi investigar búfalos do estado do Maranhão para piroplasmas e A. marginale utilizando-se a técnica da PCR. Todas as amostras foram negativas para A. marginale . Um dos 287 (0,35%) búfalos testados foi positivo para Theileria sp. O sequenciamento de um fragmento do gene 18S rDNA (356 pb) demonstrou que Theileria sp. identificado estava relacionada ao grupo T. buffeli/orientalis . Estudos futuros sobre os sinais clínicos de infecção e o principal vetor neste país são necessários.

ABSTRACT

A anaplasmose bovina é uma das principais causas de perdas produtivas e mortes no Rio Grande do Sul em rebanhos bovinos. O Anaplasma marginale é o principal agente causador da enfermidade e provoca hipertermia, anemia, prostração, abortos e perdas produtivas nos bovinos acometidos. Tendo em vista o controle deste hemoparasita em uma propriedade leiteira localizada no município de Eldorado do Sul no Rio Grande do Sul, na qual a incidência da doença era alta, 471 animais foram imunizados com Anaplasma centrale na busca de desenvolvimento cruzado para Anaplasma marginale. No experimento foi verificado que a incidência que normalmente era acima de 30% na propriedade passou para níveis inferiores a 5%. No entanto, foram verificados abortos decorrentes da imunização, principalmente nos animais que possuíam menos de 90 dias de prenhes. Já o número de mortes globais na fazenda caiu consideravelmente tendo em vista que a principal causa de morte era a anaplasmose bovina. Dos animais inoculados com A. centrale em torno de 15% apresentaram sintomatologia clínica da enfermidade e precisaram ser tratados com oxitetraciclina no período entre 15 e 30 dias após a imunização. O custo com tratamento empregado na propriedade posterior à imunização caiu em torno de 85% o que provocou impacto significativo economicamente na propriedade.(AU)


Bovine anaplasmosis is a major cause of production losses and deaths in Rio Grande do Sul cattle herds. Anaplasma sp. is the main causative agent of cattle disease. It causes hyperthermia, anemia, prostration, abortions and reduces milk production in affected animals. In order to control this hemoparasite on a dairy farm located in the municipality of Eldorado do Sul in Rio Grande do Sul, where the disease incidence was high, 471 animals were immunized with Anaplasma centrale in the search for cross-protectiv immunity for Anaplasma marginale. The property anaplasmosis incidence, which usually was above 30%, became 5% after the immunization. However, abortions were observed resulting from innoculaition, especially in animals that had less than 90 days of pregnancy. The global number of deaths on the farm dropped considerably given that the main cause of death was the bovine anaplasmosis. 15% of animals inoculated with A. centrale showed clinical symptoms of the disease between 15 and 30 days after immunization and had to be treated with oxytetracycline. The amount of money spent with anaplasmosis treatment decay 85% after the immunization, which caused significant economic impact on the property.(AU)

Abstract Vaccination against Anaplasma marginale has been considered an important control strategy for bovine anaplasmosis. Recently, mice immunized with rMSP1 a linked to carbon nanotubes (MWNT) showed significant immune responses, generating a new possibility for use of an inactivated vaccine. The objective of this study was to investigate the cellular and humoral responses in calves immunized with MWNT+rMSP1a , associated with inactivated vaccine of A. marginale produced in vitro, and evaluate the toxic effects of the MWNT on renal and hepatic function. rMSP1a was covalently linked to MWNT. Inactivated vaccine (AmUFMG2) was produced by cultivating A. marginale in IDE8 cells. Twenty-four Holstein calves were divided (four groups) and immunized subcutaneously with PBS and non-carboxylated MWNT (control, G1), AmUFMG2 (G2), MWNT+rMSP1a (G3), and AmUFMG2 with MWNT+rMSP1a (G4). Blood samples were collected for total leukocyte counts, biochemical profiling and evaluation of the cellular and humoral response. Immunization with MWNT+rMSP1a induced increase in the total number of leukocytes, NK cells, in the lymphocyte populations and higher levels of antibodies compared to calves immunized only with AmUFMG2. Furthermore, MWNT did not induce changes in the biochemical profile. These data indicate that MWNT+rMSP1a were able to induce the immune responses more efficiently than AmUFMG2 alone, without generating toxicity.


Resumo Vacinação contra Anaplasma marginale tem sido considerada uma importante estratégia de controle da anaplasmose bovina. Recentemente, camundongos imunizados com rMSP1a funcionalizada à nanotubos de carbono (MWNT) apresentaram resposta imune significante, gerando nova possibilidade para o uso da vacina inativada. O objetivo desse estudo foi investigar a resposta celular e humoral em bezerros imunizados com MWNT+rMSP1a, associado com a vacina inativada de A. marginale produzida in vitro, e avaliar os efeitos tóxicos dos MWNT nas funções hepática e renal. rMSP1 a foi ligada covalentemente aos MWNT. Vacina inativada (AmUFMG2) foi produzida através do cultivo de A. marginale em células IDE8. Vinte e quatro bezerros Holandeses foram divididos (quatro grupos) e imunizados subcutaneamente com: PBS e MWNT não-carboxilados (controle, G1), AmUFMG2 (G2), MWNT+rMSP1 a (G3), e AmUFMG2 com MWNT+rMSP1a (G4). Amostras de sangue foram coletadas para contagem de leucócitos, perfil bioquímico e avaliação da resposta celular e humoral. Imunização com MWNT+rMSP1a induziu aumento dos leucócitos totais, células NK, na população de linfócitos e altos níveis de anticorpos comparado com animais imunizados apenas com AmUFMG2. Além disso, MWNT não induziu alterações no perfil bioquímico. Esses dados indicam que MWNT+rMSP1a foram capazes de induzir eficientemente a resposta imune comparado com AmUFMG2 sozinho, sem gerar toxicidade.

Este estudo avaliou a incidência de infecções naturais pelos agentes da tristeza parasitária bovina (TPB), Anaplasma marginale, Babesia bovis e Babesia bigemina, em bezerros nascidos em cinco fazendas do semiárido paraibano. Em cada fazenda, foram coletadas amostras de sangue de 6 a 14 bezerros a cada 14 dias durante os primeiros 12 meses de vida de cada animal. As amostras de sangue foram processadas por microhematócrito e testadas por PCR para detecção de DNA de A. marginale, B. bovis e B. bigemina. Em paralelo, foram quantificadas as infestações por carrapatos nos bovinos nas cinco fazendas, assim como as populações de tabanídeos em três fazendas. De 41 bezerros monitorados durante o primeiro ano de vida, 25 (61,0%) apresentaram PCR positivo para A. marginale, 7 (17,1%) para B. bigemina e 3 (7,3%) para B. bovis. Os valores de incidência da infecção por A. marginale variaram de 83,3% a 100% em quatro fazendas. A infecção por B. bigemina ocorreu em bezerros de apenas duas fazendas (incidências de 12,5% e 85,7%) e a por B. bovis em apenas uma (incidência de 42,8%). Em uma fazenda os 14 bezerros permaneceram negativos para A. marginale, B. bigemina e B. bovis durante os 12 meses de acompanhamento. Os resultados de PCR foram confirmados por sequenciamento de DNA de produtos amplificados. A presença de carrapatos Rhipicephalus (Boophilus) microplus foi verificada somente em duas propriedades, nas quais houve infecção por A. marginale, B. bigemina e B. bovis (este último agente em apenas uma delas). Foram capturados 930 tabanídeos no estudo, a maioria durante os períodos de chuvas na região; 70,7% dos tabanídeos corresponderam a Tabanus claripennis. Houve associação significativa entre PCR positivo para A. marginale ou B. bigemina e menores valores de hematócrito. Este estudo demonstra que, mesmo avaliando apenas cinco propriedades rurais, a incidência dos agentes da TPB ocorreu de forma heterogênea na região, corroborando o status de área de instabilidade enzoótica para TPB previamente relatado para o semiárido paraibano.(AU)


This study evaluated the incidence of natural infection by agents of cattle tick fever (CTF), Anaplasma marginale, Babesia bovis and Babesia bigemina in calves born in five farms within the semiarid region of Paraíba state, Brazil. In each farm, blood samples were collected from 6 to 14 calves every 14 days during the first 12 months of life of each animal. Blood samples were processed by microhematocrit and tested by PCR for detection of DNA of A. marginale, B. bovis and B. bigemina. In parallel, the tick infestations on animals were quantified in the five farms, as well as populations in horseflies in three farms. From a total of 41 calves monitored during the first year of life, 25 (61.0%) had positive PCR for A. marginale, 7 (17.1%) for B. bigemina and 3 (7.3%) to B. bovis. Incidence values for A. marginale infection ranged from 83.3% to 100% in four farms. Infection with B. bigemina in calves was detected at only two farms (incidence of 12.5% and 85.7%) and by B. bovis in just one (42.8% incidence). On one farm 14 calves remained negative for A. marginale, B. bigemina and B. bovis during the 12 month follow-up. PCR results were confirmed by DNA sequencing of amplified products. The presence of Rhipicephalus (Boophilus) microplus was found only in two farms in which there was infection by A. marginale, B. bigemina and B. bovis (the latter agent in only one of them). A total of 930 horseflies were captured in the study, most during periods of rain in the region; 70.7% of horseflies corresponded to Tabanus claripennis. There was significant association between a positive PCR for A. marginale and B. bigemina and lower hematocrit values. This study demonstrates that even evaluating only five rural properties, the incidence of CTF occurred heterogeneously in the region, confirming the status of enzootic instability area for CTF, previously reported for the semiarid region of Paraiba.(AU)

La anaplasmosis es una enfermedad infecciosa que afecta a los bovinos y es causada por la rickettsia intraeritrocítica Anaplasma marginale. A pesar de su impacto global, hasta el momento, no se ha desarrollado una vacuna efectiva. Se ha postulado que la inmunización del ganado con la membrana externa de la rickettsia, genera protección parcial contra la exposición al Anaplasma marginale y que esta respuesta está dirigida a seis polipéptidos denominados Proteínas Principales de Superficie (MSPs del inglés, Major Surface Proteíns). Estas proteínas presentan un carácter inmunodominante cuando se emplean como antígenos en su forma nativa o recombinante, lo que ha hecho pensar que son los antígenos indicados para una vacuna. Sin embargo, la protección brindada fluctúa entre una protección total o parcial a ninguna protección. La búsqueda de antígenos alternativos a las MSPs, se ha visto limitada a consecuencia de su muy baja o ausente capacidad inductora de formación de anticuerpos o de linfocitos T reactivos, los cuales son indispensables para su identificación mediante inmunoensayos. El objetivo de este trabajo fue describir las estrategias aplicadas recientemente para identificar nuevos antígenos de A. marginale, denominados antígenos subdominantes, porque representan una pequeña porción del proteoma de la rickettsia, los cuales han demostrado ser buenos candidatos vacunales contra la anaplasmosis. Actualmente, diversos estudios moleculares y bioinformáticos han definido varias proteínas de superficie y componentes del Sistema de Secreción Tipo IV, como antígenos potenciales; entre ellos están las proteínas Ana17, Ana29, Ana32, Ana37 y Ana43, que le han brindado una protección parcial al ganado después de la exposición al proceso infeccioso; y las proteínas VirB9-1, VirB9-2 y VirB10 (Sistema de Secreción tipo IV), las cuales generan una fuerte respuesta inmune en el ganado, aunque se desconoce su nivel de protección.


The Anaplasmosis is an infectious disease of cattle caused by the intraerythrocytic rickettsia Anaplasma marginale. Despite its overall impact, no effective vaccine has been developed so far. It has been postulated that immunization of cattle with the native outer membrane of rickettsia generates partial protection against Anaplasma marginale challenge; and that this response is directed to six polypeptides known as Major Surface Proteins (MSPs). These proteins display an immunodominant character when they are used as immunogens in their native or recombinant form, which has suggested that they are the appropriate vaccine antigens. However, the protection provided varies from a total or partial protection to no protection. The search for alternative antigens to MSPs has been limited as a result of their very low or null capacity for inducing the production of potential antibodies or reactive T cells, which are essential for their identification through immunoassays. The objective of this review was to describe the recently applied strategies for the identification of new antigens of A. marginale, the so-called subdominant antigens, proteins that represent a small portion of the proteome of the rickettsia. These proteins have shown to be good vaccine candidates against anaplasmosis. Currently, molecular and bioinformatics studies have defined several surface proteins and components of the Type IV Secretion System as potential antigens; among them are the Ana17, Ana29, Ana32, Ana37, and Ana43 proteins, which have given partial protection to cattle after the infectious challenge; and VirB9-1, VirB9-2, and VirB10 proteins (Type IV Secretion System), which generate a strong immune response in cattle, although their level of protection is unknown.

This study aimed to determine the seroprevalence of Babesiosis and Anaplasmosis in cattle from the municipalities of Ouricuri and Petrolina, state of Pernambuco, Brazil, and to define the risk factors for the occurrence of the diseases. Blood samples were collected for serologic testing by Indirect Immunofluorescence Assay (IFA). Sanitary epidemiological questionnaires were applied to the producers aiming to identify possible risk factors. Ticks were collected, identified and tested by Polymerase Chain Reaction (PCR) for the diagnosis of infection by Anaplasma marginale, Babesia bigemina and Babesia bovis. The study was conducted with 861 cattle, being 468 in Petrolina and 393 in Ouricuri. The seroprevalence of A. marginale, B. bigemina and B. bovis in Petrolina was of 35.0% (164/468), 35.9% (168/468) and 32.3% (151/468), respectively; and in Ouricuri was 45.5% (179/393), 38.6% (152/393), and 54.9% (216/393), respectively. Co-infection for Anaplasma spp. and Babesia spp. was observed in 31.6% and 32.1% samples of Petrolina and Ouricuri, respectively. The detection of DNA of Babesia spp. by PCR was possible in 5.8% (8/137) ticks; which 62.5% (5/8) was detected later infection with B. bovis; and 23.3% (32/137) with A. marginale. The presence of ticks, the use of acaricide, age, race, and county of residence of the animals were identified as risk factors for TBD by univariate analysis and multivariate. This study allowed the characterization of the municipalities studied as enzootic instability areas for these hemoparasitic, and consequently alert for adoption of adequate control measures and new studies.

ABSTRACT

Abstract The objective of this study was to assess the occurence of animals seropositive for Anaplasma marginale in the municipality of Realeza, Paraná State, Brazil. Blood samples were collected from 344 cows on 18 small farms in the municipality of Realeza-PR. The animals’serum samples were forwarded to the Federal University of Fronteira do Sul, in order to investigate the occurrence of anti-A. marginale IgG antibodies by an enzyme-linked immunosorbent assay commercial kit. IgG antibodies to A. marginale were detected in cattle from 77.7% of the farms. To the best author's knowledge, this is the first report of occurrence of A. marginale in cattle in southwestern Paraná. The serological assay showed that 24.4% of the animals were seropositive, thus characterizing the location investigated as an area of enzootic instability for the disease. The family farms located in the municipality of Realeza-PR showed enzootic instability for bovine anaplasmosis. It is necessary to conduct disease monitoring programs in association with preventive measures in order to ensure the sanitary quality of the herds and to reduce economic losses for the farmers. In addition, it is essential to implement educational extension actions that allow farmers to acquire knowledge, attitudes and perceptions regarding the risk factors that contribute towards herd A. marginale-infection.


Resumo O objetivo deste estudo foi avaliar a ocorrência de animais soropositivos para Anaplasma marginale, no município de Realeza, Estado do Paraná, Brasil. Foram colhidas amostras de sangue de 344 fêmeas bovinas provenientes de 18 propriedades rurais do município de Realeza - PR. Amostras de soro dos animais foram encaminhadas à Universidade Federal da Fronteira Sul, para realização do Ensaio Imunoenzimático Indireto para pesquisa de anticorpos IgG anti- Anaplasma marginale por meio de kit comercial. Anticorpos IG anti-A. marginale foram detectados em 77,7% das propriedades. Trata-se do primeiro registro da ocorrência de A. marginale no Sudoeste paranaense. A sorologia evidenciou 24,4% de animais soropositivos, caracterizando o local pesquisado como área de instabilidade enzoótica para a doença. As propriedades de agricultura familiar, localizadas no município de Realeza-PR, apresentaram instabilidade enzoótica para anaplasmose bovina. É necessário que programas de monitoramento da enfermidade sejam realizados em conjunto com medidas de prevenção, visando garantir qualidade sanitária do rebanho e reduzir perdas econômicas dos produtores rurais. Além disso, é fundamental a realização de ações de extensão que viabilizem a aquisição de conhecimento, atitude e percepção dos criadores diante dos fatores de risco contribuintes para a infecção por A. marginale do rebanho.

ABSTRACT

In February 2012, a reproductive group of 60 adult blackbucks (Antilope cervicapra) from Veracruz, Mexico was relocated to hunting units in eastern and northeastern Mexico. Seven individuals died due to hemorrhagic parasitic, abomasitis and enteritis caused by Haemonchus spp., Setaria spp., and Trichostrongylids. Deaths were associated with hepatic necrosis, bilateral congestive distention of heart and fibrinonecrotic bronchopneumonia. Also Anaplasma marginale was identified. The blackbucks' population displayed a general mortality rate of 11.67%, where 25% of total male and 9.62% of total female died. The mortality was controlled by segregation of all remaining blackbucks and the treatment for internal and external parasites (biting flies and ticks). After the treatment, no fatality cases related to parasitosis were recorded. The results presented here exhibit the high relevance of parasitosis as possible factor risk in the survival of tis specie.


En febrero del 2012, un grupo de 60 individuos adultos reproductivos de antílope negro (Antilope cervicapra) provenientes de Veracruz, México fueron reubicado en unidades de cacería del este y noreste de este país. Siete individuos murieron presentando hemorragias parasíticas, abomasitis y enteritis ocasionadas por Haemonchus spp., Setaria spp. y Trichostrongylids. Las muertes estuvieron asociadas con necrosis hepática, distensión congestiva del corazón y bronconeumonía fibronecrótica, donde Anaplasma marginale fue identificada. La población de antílopes negros mostró un porcentaje de mortalidad del 11.67%, en donde el 25 y 9.62% de los machos y hembras totales murieron. La mortalidad fue controlada mediante el aislamiento de los antílopes negros restantes y un tratamiento contra parásitos internos y externos (garrapatas y moscas picadoras), lo cual controló las mortalidades y reveló la importancia de la parasitosis como factor de riesgo que afecta la sobrevivencia de esta especie.

ABSTRACT

Anaplasma marginale (A. marginale), es una bacteria del orden de las Rickettsias que ocasiona la anaplasmosis bovina en regiones tropicales y subtropicales del mundo. Esta enfermedad, trasmitida principalmente por tábanos y garrapatas, se desarrolla típicamente en una etapa inicial aguda con manifestaciones clínicas caracterizadas principalmente por anemia y fiebre. Después de un par de meses, los animales recuperan su condición física y se hacen asintomáticos, siendo incapaces de eliminar completamente la bacteria, convirtiéndose en animales persistentemente infectados. Esto se debe a la capacidad de A. marginale para evadir el sistema inmune. En este sentido, se ha demostrado la existencia de un mecanismo de variación antigénica en las proteínas MSP1, MSP2 y MSP3 de la bacteria. Al evaluar la familia multigénica que codifica para la MSP2, se determinó que está conformada por dos regiones conservadas que flanquean una región central hipervariable. De esta manera, al expresarse cada una de las 52 variables de la MSP2, se expresa un epítope diferente. Cuando se describió el genoma completo de este hemotrópico, se encontró también la presencia de 16 pseudogenes msp2, los cuales pueden ser recombinados dentro del sitio de expresión del operón de la MSP2, constituyendo un segundo mecanismo de variación. Además de ello, los fragmentos hipervaribles y los pseudogenes se pueden combinar entre sí, en un proceso denominado conversión génica, creando nuevos epítopes “recombinantes”, confiriendo una capacidad de variabilidad antigénica casi infinita al A. marginale (tercer mecanismo). Un cuarto mecanismo de variación antigénica, lo constituye la dimerización de la MSP2 sobre la superficie del A. marginale, debido a que la expresión simultánea de variantes conforman epítopes únicos. En conclusión, la recombinación génica de la MSP2 y su dimerización en la membrana, constituye un mecanismo muy eficiente de variación antigénica para eludir el sistema inmunológico del hospedador.


Anaplasma marginale (A. marginale) is a bacterium of the Rickettsiales order that causes bovine anaplasmosis in tropical and subtropical regions worldwide. This disease, mainly transmitted by ticks and horseflies, typically develops in an initial acute stage, with clinical signs characterized by anemia and fever. After two months, animals recover their original physical condition and become asymptomatic, being unable to completely eliminate the bacterium, turning into persistently infected animals. This is due to the ability of A.marginale to evade the immune system. In this regard, the existence of a mechanism for antigenic variation in proteins of the bacterium, such as MSP1, MSP2, and MSP3, has been demonstrated. When assessing the multigenic family which encodes for MSP2, it was determined that it consists of two conserved regions flanking a central hypervariable region. Thus, when expressing each of the 52 MSP2 variables, a different epitope is also expressed. When the entire genome of this parasite was decoded, the presence of 16 pseudogenes for MSP2 was also discovered. These pseudogenes can be recombined within the operon expression site of MSP2, providing a second mechanism of variation. Moreover, both the hypervariable fragments and pseudogenes can combine among them, in a process called gene conversion, creating new “recombinant” epitopes, conferring the A. marginale with an almost infinite capacity for antigenic variability (third mechanism). A fourth mechanism of antigentic variation consists of the dimerization of MSP2 on the surface of A. marginale, because the simultaneous expression of variants creates unique epitopes. In conclusion, gene recombination of MSP2 along with the dimerization of MSP2 on the membrane provides a very efficient mechanism for antigenic variation for evading the host’s immune system.

ABSTRACT

O objetivo do estudo foi testar a prevalência sorológica e molecular de Anaplasma marginale em búfalos do municipio de Soure, Ilha de Marajó, estado do Pará, Brasil. Para a pesquisa sorologica foram selecionados randomicamente 800 animais e para a pesquisa molecular 50 destes animais foram aleatoriamente escolhidos. Para quantificar a prevalência sorológica utilizou-se o ensaio de imunoadsorção enzimático indireto (iELISA) com antígeno total contendo proteínas de superfície externa e para quantificar a prevalência molecular utilizou-se a reação em cadeia da polimerase (PCR), envolvendo a amplificação de fragmento gênico da proteína de superfície maior 5 (MSP5). A prevalência de animais positivos no ELISA para A. marginale foi de 25% (200/800). Na PCR foi detectada a presença de A. marginale em 2% (1/50) dos animais. Embora apenas um animal tenha sido positivo na PCR, observou-se que o mesmo foi negativo no ELISA. A presença do agente, mesmo em baixa prevalência, mostra que os bubalinos podem funcionar como um importante reservatório desse patógeno para os rebanhos bovinos da região norte do Brasil.


The aim of the study was to test the molecular and serological prevalence of Anaplasma marginale in water buffaloes of the Marajó Island, State of Pará, Brazil. For serologic research were randomly selected 800 buffaloes and for molecular research 50 of these animals were randomly chosen. To quantify the serological prevalence we used the indirect enzyme linked immunosorbent assay (iELISA) with total antigen containing proteins outer surface. To quantify the prevalence molecular was used the polymerase chain reaction (PCR) involving gene amplification fragment larger surface protein 5 (MSP5). The prevalence of positive animals in iELISA was 25% (200/800). In the PCR we detected the presence of A. marginale in 2% (1/50) of animals. Although only one animal was positive in PCR, we found that it was negative in ELISA. The presence of the agent, even in low prevalence, shows that buffaloes can act as an important reservoir for transmission of the pathogen to cattle in northern Brazil.

ABSTRACT

The rickettsia Anaplasma marginale is considered the main agent of bovine anaplasmosis. Due the nonspecific clinical signs of the anaplasmosis, the diagnosis of infection depends of laboratory confirmation. In recent years, molecular diagnostic methods have been used to detect A. marginale in cattle. However, the existence of a large number of assays of different sensitivity and cost makes the choice of an appropriate test difficult. In the present study, a real-time Polymerase Chain Reaction (PCR) based on the msp5 target gene was quantitatively assessed and compared to an end point PCR. Both reactions were subjected to sensitivity and specificity evaluation using plasmid DNA and samples from cattle experimentally infected with A. marginale. A comparative field trial of the tests was carried out using samples of cattle from a stable enzootic area for A. marginale. The real-time PCR showed a higher sensitivity than the end point PCR. This reaction (i.e. real-time PCR) was able to detect one copy of the msp5 gene in 100 ηg of plasmidial DNA, and more than 80% of its results were positive among experimentally infected animals seven days after infection. In addition, based on in silico analysis, the real-time PCR evaluated in the present study appears to be useful for the detection of A. ovis.


A riquétsia Anaplasma marginale é considerada o principal agente da anaplasmose bovina. Devido a não especificidade dos sinais clínicos, a confirmação da infecção nos animais depende de testes laboratoriais. Recentemente, métodos de diagnóstico molecular têm sido aplicados para detecção de A. marginale em bovinos. No entanto, a grande quantidade de testes com diferentes sensibilidade e custos tem dificultado a escolha do ensaio mais adequado. No presente estudo, uma PCR em tempo real baseada no gene msp5 foi avaliada quantitativamente e comparada a uma reação de PCR convencional. As reações foram submetidas à avaliação de sensibilidade e especificidade com DNA plasmidial e amostras provenientes de bovinos experimentalmente infectados por A. marginale. Uma avaliação comparativa a campo foi realizada entre os testes utilizando amostras provenientes de bovinos criados em uma região de estabilidade enzoótica para A. marginale. Embora os testes não tenham apresentado diferença estatisticamente significativa, a PCR em tempo real apresentou valor de sensibilidade maior do que a PCR convencional. A PCR em tempo real foi capaz de detectar uma cópia de msp5 em 100ng de DNA plasmidial, e mais de 80% de resultados positivos entre bovinos experimentalmente infectados apenas sete dias após infecção. Além disso, baseado em análise in silico, a PCR em tempo real avaliada aqui pode ser útil para detecção de Anaplasma ovis.

ABSTRACT

The recombinant protein MSP5 has been established as an important antigen for serological diagnosis of Anaplasma marginale by enzyme-linked immunosorbent assay (ELISA). However, due to the high cost of specialized equipment, this technique is not accessible to all laboratories, especially in developing countries in areas where the disease is endemic. The present study describes the standardization of a latex agglutination test (LAT) to detect antibodies against A. marginale based on recombinant MSP5. Compared with indirect enzyme-linked immunosorbent assay (iELISA), the relative sensitivity and specificity of the LAT were 95.21% and 91.86% respectively, with an almost perfect agreement between tests (kappa index = 0.863). These results can be considered important for the serological diagnosis of A. marginale, as they indicate that the test represents a rapid and low cost alternative to ELISA.

ABSTRACT

Outbreaks of tick-borne disease cases in Santa Catarina, Brazil are known, but the presence of the pathogen DNA has never been determined. In this study, the first survey of Anaplasma marginale, Babesia bigemina, and Babesia bovis DNA on blood samples of 33 cattle from an outbreak in Ponte Alta Municipality, Santa Catarina, Brazil, has been carried out. A multiplex PCR detected 54.5% of animals were co-infected with 2 or 3 parasites, while 24.2% were infected with only 1 species. The most prevalent agent was B. bigemina (63.6%) followed by A. marginale (60.6%). This is the first report of tick-borne disease pathogens obtained by DNA analysis in Southern Brazil.
